E2F7 binds to the FOXO1 promoter and restrains its expression. The E2F7 binding site in the FOXO1 promoter was analyzed using bioinformatics methods (A). ChIP experiment (B) and dual luciferase reporter assay (C) confirmed the binding of E2F7 to the FOXO1 promoter. The expression of FOXO1 mRNA and protein in MCF7 and MDA-MB-231 cells after knockdown or overexpression of E2F7 was assessed using RT-qPCR (D) and western blot assay (E).
