Effect of simvastatin on DNA repair gene expression in prostate cancer and prostate stromal cells (PrSC). PC-3, 22Rv1, LNCaP-LA, LNCaP, and PrSC cells were incubated with the medium containing 10% FBS for 24 h, and the medium was switched to the indicated concentration of simvastatin in the medium containing 10% FBS. After 48 h, the total RNA (A, C) and total protein (B, D) were collected. (A) Comparison of mRNA expression levels of each gene in each cell without simvastatin. mRNA expression of BRCA1, BRCA2, RAD51, FANCD2, FANCG, FANCA, BARD1, RFC3, RFC4, and RFC5 was evaluated by performing real-time polymerase chain reaction tests, and the relative quantitative volume (RQV) was calculated by comparing the expression of β-actin.
